「Alzheimer’s Disease:Diagnostic Complexity and Emerging Therapeutics 」

This presentation will provide an overview of dementia as a progressive syndrome affecting over 55 million people worldwide, with Alzheimer’s disease (AD) being the most prevalent cause. We will explore the distinction between reversible and irreversible causes, the clinical features of neurocognitive disorders, and hallmark brain changes in AD such as beta-amyloid plaques and tau tangles. Approved symptomatic treatments and the first generation of anti-amyloid therapies will be discussed, along with disease-modifying treatments under investigation.  The complexity of mixed dementia, marked by coexisting pathologies will be presented to underscore the need for future drug development to adopt a precision medicine approach for more effective and individualized interventions.

本講演では、世界で5,500万人以上が罹患する進行性症候群である認知症について概説し、その中でも最も一般的な原因であるアルツハイマー病（AD）に焦点を当てる。可逆的および不可逆的な原因の区別、神経認知障害の臨床的特徴、そしてベータアミロイドプラークやタウタングルなどのADにおける特徴的な脳内変化について探る。承認された対症療法や第一世代の抗アミロイド療法について議論するとともに、現在研究中の疾患修飾治療についても取り上げる。複数の病理が共存する混合型認知症の複雑性についても紹介し、より効果的で個別化された介入のために、将来の薬剤開発において精密医療アプローチを採用する必要性を強調する。
